由于增加的轴距全部贡献给了车身后部，因此在后门的设计上，梅赛德斯-迈巴赫S级将车门的三角窗设计在了C柱上，车门呈现出更为传统的方形。在车内营造出了双C柱的视觉感受，提升了后排的私密感与安全感，同时这一设计也遵从了迈巴赫传统的后门设计，呈现出了设计上的尊贵感。

梅赛德斯-迈巴赫S600后排的座椅是迄今最舒适的。后排提供了两个独立的行政座椅，左右两侧拥有了相同的尊贵享受。座椅可以实现19~43.5°的角度调节，而43.5°的角度，几乎是一种半躺的姿势，这也是能带给乘客最舒适乘坐感受的座椅角度。即便是在如此半躺的坐姿时，后排仍可以有着足够的腿部空间，比任何竞争对都更有优势。

与常规的奔驰S级相同，梅赛德斯-迈巴赫S600后排座椅也提供了柔软的头枕，腿部支撑也拥有了一个柔软的靠垫。此外座椅按摩功能采用了热石原理，将14个独立传动的气垫加在靠背中，具备了加热功能、放松效果、促进血液循环等功能。

当你坐进这台梅赛德斯-迈巴赫S600后排座椅后，原本萦绕在你耳边的噪音，会一下子静了下来，提供给用户一个静谧的乘坐环境。在其车窗玻璃使用了是三层材料，车门内侧的密封条使用了双层材料，对于习惯乘坐后排的乘客，梅赛德斯-迈巴赫S600成为了全球后排空间最安静的量产轿车。

当然梅赛德斯-迈巴赫S600的舒适乘坐感受，也少不了智能车身控制系统（俗称：魔术底盘）在行驶中带来的效果。它通过位于风挡玻璃后方的立体摄像头，可对前方路面的坑坑坎坎扫描，并通过电脑进行计算，对减震器进行调节，减缓行驶中路面的颠簸感。有了这套装备，遇到减速坎，你不必在放慢车速了，正常行驶通过，实际车内的感受却非常细微，车身依旧能保持平稳，不受影响。此外，主动悬挂还可有效抵消起步、转弯或制动时的车身倾翻和颠簸。

梅赛德斯-迈巴赫S600，车内仪表台上提供了一个IWC万国模拟时钟，它在全车中也成为梅赛德斯-迈巴赫S600品质精湛、可靠的象征。

座椅使用了designo 尊贵版诗米安(半苯胺)皮革，这种皮革在加工处理上，需要多达60的加工时间，其表面未经打磨，通风性极佳，最大程度还原了真皮的细腻质感。也最大限度保持原始的色彩，不进行过多的喷涂处理。装饰纽扣和布达佩斯滚边处理以及菱形绗缝图案都提升了它的精致、尊贵以及独特感。车内其余部分的皮质，都使用了高档的Napa皮革进行包裹。作为豪华商务价值的体现，梅赛德斯-迈巴赫S600为后排乘客提供了可以折叠的小桌板，纯金属的支架，呈现出来工艺的高贵与精致。

Burmester®高端3D环绕立体声音响，也成为了给梅赛德斯-迈巴赫S600增色的奢华装备，在梅赛德斯-迈巴赫S级轿车中属于格外尖端的设备。它配备了24个高性能音箱和24条独立功放声道，总输出为1,540瓦。当主机打开时，它们会360度旋转，以螺旋运动方式伸出10毫米。这一调整可使高音音箱对齐，来提供最佳声音体验。

价值11.2万元的自动调光的全景天窗在这台梅赛德斯-迈巴赫S600上，是唯一作为选装项目，提供给用户功能配置。在夏季可使车内温度最多降低 10°C。当拔下车钥匙且车顶断电时，玻璃颜色会在车辆停驻后自动变暗，从而防止车内升温并防止车内被窥视。
